About Celecoxib Non-steroidal anti inflammatory drug (NSAID) ,COX-2 Selective inhibitor, Anti-inflammatory Agent. Mechanism of Action of Celecoxib This inhibits prostaglandin synthesis peripherally by selective noncompetitive inhibition of cyclooxygenase-2 (COX-2) enzyme. It binds with its polar sulfonamide side chain to a hydrophilic side pocket region close to the active COX-2 binding site (Both COX-1 and COX-2 catalyze the conversion of arachidonic acid to prostaglandin (PG) H2, the precursor of PGs and thromboxane). This results its anti-inflammatory action Indications for Celecoxib 1. Osteoarthritis 2. Rheumatoid arthritis 3. Acute gout Contra-indications of Celecoxib 1. Hypersensitivity to celecoxib 2. Allergic type reactions to sulphonamides /aspirin or other NSAIDs.
